C/EBPβ siRNA reduces C/EBPβ expression and inhibits dexamethasone-induced C/EBPβ mRNA and protein levels in L6 myoblasts. (A) C/EBPβ mRNA levels in L6 myoblasts transfected with C/EBPβ siRNA or non-targeting siRNA and treated for 24 h with or without 1 µM dexamethasone. C/EBPβ mRNA in myoblasts transfected with non-targeting siRNA and cultured for 24 h in the absence of dexamethasone was arbitrarily set at 1.0. Results are means ± SEM with n=6 per group. *p<0.05 vs –Dex; +p<0.05 vs corresponding non-targeting group by ANOVA. (B) C/EBPδ mRNA levels in the same groups of myoblasts as depicted in panel A. Molecular weights on the right side of the blots were based on molecular weight markers used in the Western blots. Almost identical results were observed in three repeated experiments.
