Presence of viral RNA in ticks after infection. Total RNA was isolated from a group of 25 Ixodes scapularis larvae at 21 days postimmersion with medium only (mock) or the various viruses. Viral RNA was detected using primers specific for the (A) positive- or (B) negative-sense RNA. To detect TBEV/DEN4Δ30 and its derivatives (vΔ30/E₃₁₅, vΔ30/NS5_654,655, and vΔ30/E₃₁₅/NS5_654,655), TBEV-specific primers were used. LGT- or DEN4-specific primers were used to detect LGT/DEN4, LGT, or DEN4. Total RNA isolated from virus infections in Vero cells were used as positive controls (+CNTRL).
