(A) To increase the solubility of 6a two derivatives 7a and 7b were synthesized. In vitro-activated Hog1 as was inhibited 7a and 7b. (B) hog1as cells transformed with the STL1:LacZ reporter were pre-incubated with 2 µM of 7a and 7b for the indicated times. Cells were subjected to osmotic stress (0.4 M NaCl) for 30 minutes and gene expression measured as before. β-galactosidase activity is given as fold induction of control versus NaCl-treated cells (activity given in nanomols per minute per milligram).
