Inhibition of p38 increases HSC long-term engraftment after primary transplantation. LKS+ cells were cultured ex vivo in the presence of vehicle (0.1% DMSO) or SB (5 μM) for 7 days. The expanded cells harvested from the culture with 200 LSK+ cells were transplanted into a lethally irradiated mouse along with 2×105 competitors. Donor cell engraftment in peripheral blood was determined 8 and 20 weeks after transplantation. (A, C) The data are presented as mean±SE (n=8 mice/group) of percent of total donor cell engraftment. (B, D) The data are presented as mean±SE (n=8 mice/group) of percent of donor myeloid, T and B cell engraftment. ***P<0.001.
