Electrophoretic mobility shift assay. A radioactively labeled anti-AB or AB RNA (500 nM) was incubated with increasing concentrations of PFV PR-RT in 50 mM Na₂HPO₄-NaH₂PO₄ (pH 6.4), 100 mM NaCl, 10% glycerol at room temperature for 5 min. For displacement, increasing concentrations of unlabeled anti-AB or AB RNA were added to a mixture containing 500 nM radioactively labeled AB RNA and 2 μM PFV PR-RT as indicated above the gels. Complexes were separated on 6% DNA retardation gels at 4°C. Numbers below the gels show the remaining free RNA in percentages (including the standard deviations), as quantitated by densitometry.
