Increased hexokinase activity inhibits apoptosis in a glucose dependent manner. Ad-LacZ or Ad-HKI infected Rat1a cells were grown in the presence (A,B) or absence (C,D) of glucose. The total hexokinase activity of the infected cells was 31.5 (±4.5) U/g for Ad-LacZ and 50.5 (±7.5) U/g for Ad-HKI. Apoptosis was induced by growth factor withdrawal/ultraviolet, and at the indicated times, cells were fixed and stained with anti-cytochrome c antibodies or DAPI. The percentage of cells with released cytochrome c (A,C) and condensed chromatin (B,D) are shown (P < 0.002). The results are shown as average (±SE) of four independent experiments.
