Cdc9p is required for mitochondrial DNA maintenance. Yeast strains were grown in glucose to mid-log (A) or to stationary (B) phase at the permissive temperature and subsequently shifted to the non-permissive temperature. At the times indicated genomic DNA was harvested and mitochondrial DNA was examined. Strain 2780-49BWT, cdc9-1^ts yeast, cdc9-1^ts yeast harboring pYES2-CDC9 and cdc9-1^ts yeast harboring pYES2-N-CDC9 were analyzed. The upper panels depict Southern blot hybridizations to a mitochondrial DNA probe, the lower panels depict the corresponding ethidium bromide stained agarose gels. The molecular weight marker is HindIII-digested phage λ DNA.
