Caspase activity in strains of C. albicans. Cells of each strain were grown overnight, standardized, and inoculated in YPD medium in the absence (A) or presence (B) of 10 mM H₂O₂. Caspase activity was measured using a poly-caspase (FAM-VAD-FMK) apoptosis detection kit (Immunochemistry Technologies, LLC). At designated times, FAM-VAD-FMK was added to cell suspensions. After unbound FAM-VAD-FMK was washed away, anti-FAM-VAD-FMK antibody was added. The number of cells with green fluorescence is a direct measurement of caspase-positive cells.
