Transient receptor potential melastatin type 7 channel (TRPM7) knockdown induces mesenchymal stem cell death. (A) TRPM7 gene expression 7 days after lentivirus infection with nonspecific shRNA (control) or TRPM7 shRNA and puromycin selection. The qPCR data represents the ratio of TRPM7 to the housekeeping gene GAPDH. (B) Cells from control and TRPM7 shRNA groups (2.5 × 105) were seeded in 2 different culture flasks on day 7 post-infection and allowed to grow for 14 days in expansion medium. Note the decrease in MSC population in knockdown cells. (C) The effect of TRPM7 knockdown on mesenchymal stem cells (MSCs) survival was examined by MTT assay. A significant reduction in cell proliferation and viability was observed in TRPM7 shRNA cells compared to controls in all time points. Data are shown as means ± s.e.m.
