Rear polarization of the MTOC in migrating neointimal SMCs involves ARPC5- and RHAMM-dependent mechanisms. PKCα phosphorylates and activates ARPC5 and RHAMM. ARPC5 coordinates the organization of the actin net above the nucleus. RHAMM coordinates microtubule-based motors to orient the MTOC rearward of the nucleus. Actin filaments emanating from the nodes of the actin net regulate both the position of the nucleus relative to the cell edge and the advancing lamellipodia during migration.
