Repression of the c-fos promoter in HSP70 overexpressing cells is blocked by cotransfection with antisense oligonucleotides directed against HSP70 mRNA. Increasing concentrations of HSP70 antisense oligonucleotide (μM) and sense oligoneucleotide (μM) were cotransfected with constant dosage of Ras (100 ng/well), HSP70 (50 ng/well) expression plasmid, and c-fos reporter in CHO-K1 cells. Cellular luciferase activity was assayed as described in the Materials and Methods section. Anti: HSP70 antisense oligos; Sense: HSP70 sense oligos. Cells were harvested 16 to 20 hours after transfection, and whole-cell extracts were subjected to luciferase assay.
