Catalytic activity of various ALOX15 Q287 mutants. Wild-type and mutant enzymes were expressed and purified as described under “Materials and Methods.” LOX concentrations of each preparation were quantified by immunoblotting, and equal amounts of enzyme were used in the activity assays. The y axis was normalized to the product amount formed by the N287Q mutant. Note that in comparison with Table 2, the amount of enzyme used was increased 3-fold.
