(A) Target site additive effect improves translational repression. Untreated RRL was programmed with Luc and Luc-451 bearing 2, 4 or 6 target sites mRNAs (designated, Luc-451X2, X4 and X6) and luciferase activity was measured 30 min after the beginning of translation. Error bars correspond to the standard deviation calculated from three independent experiments. (B) Untreated RRL was programmed with 0.46, 4.6 or 46 fmol of Luc and Luc-451X6 mRNAs and luciferase activity with Luc-451X6 was measured as a percentage of that observed with the same concentration of Luc mRNA after 30 min of translation. The molar ratio of miR-451 to the target mRNA is indicated on the bottom of the figure. Error bars correspond to the standard deviation calculated from three independent experiments.
