Western blot analysis with an AHR specific antibody was performed to confirm protein expression. Cell strains expressing TAP-tagged GFP (GFP-TAP) or murine AHR (AHR-TAP) were established in AHR null (−/−) MEFs. AHR +/+ (WT) MEFs are included as a positive control (A). GFP-TAP and AHR-TAP were also expressed in Hepa1c1c7 cells. The parental Hepa1c1c7 cells are included as a negative control (CNT). β-actin was used as loading control (B). Functional activity of AHR-TAP (C) was determined by qRT-PCR of Cyp1A1 induction in wild type (WT), AHR −/− (−/−), AHR −/− expressing AHR-TAP, and AHR −/− expressing GFP-TAP MEFs. Cell strains were exposed to DMSO (0.01% vehicle control) and TCDD (10nM) for 6 h, RNA was isolated and quantitated for Cyp1A1 levels using SYBR green. Cyp1A1 levels were normalized to HPRT. * = p < 0.05 when compared to DMSO treated cells within the same cell line.
