pH-Induced Change in Glycolipid Transfer Activity of HET-C2. Förster energy transfer involving fluorescently-labeled glycolipid (AV-GalCer) and phospholipid (Perylenoyl-PC) was used to assess glycolipid intervesicular transfer by HET-C2. Stock protein at pH 8 was diluted ~10-fold into buffer of the specified pH and incubated at room temperature for 10 min. Assays were initiated by ~1000-fold dilution of HET-C2 into indicated pH. AV-GalCer was excited at 370 nm and emission was monitored at 425 nm. Additional details are provided in the Experimental Procedures.
