(A–B) Accumulation of PAR was found in many cells in P23H and especially in S334ter retinas. (C) PAR WB identified a band at ∼116 kDa which most likely represented PARP-1 itself and additionally strong labelling of proteins ranging from 100 to 240 kDa, suggesting poly(ADP-ribosyl)ation. Retinal PAR levels were increased in both transgenic animals. (C–D) PARP-1 protein expression was similar in wt and P23H retina, but significantly increased in S334ter. PARP-1 knock-out retina was used as negative control. Values are mean ± SD from four different experiments each containing retinas from 6 animals. ***P<0.001. Scale bar = 50 µm.
