Mice infected as pups or adults have different proportions of B-cell subsets in the spleen following injection with CpG. Eight-day-old mice were infected with γHV-68 and at 30 dpi were injected with 10 nmol CpG-ODN. After 3 d the spleens were removed and put into a single-cell suspension. The cells were stained with either FITC-PNA and PE-anti B220, or PerCP/Cy5.5-CD19, PE/Cy7-CD23, and APC-CD21, to determine the B-cell phenotype.
