MK-STYX knockdown phenocopies Bax/Bak and caspase 9 knockdown. (A and B) HeLa cells were transfected with control or MK-STYX, Bax plus Bak1, caspase 8, or caspase 9 siRNA for 24 h. The cells were plated onto a 96-well e-plate and allowed to adhere and proliferate for 24 h. The cells were treated with either 10 nM paclitaxel (A) or 1 μg/ml TNF-α and cycloheximide (B) to induce intrinsic or extrinsic apoptosis, respectively. Cell viability was tracked in real time using the xCelligence system. The error bars indicate standard deviations.
