The flagella of CSC amiRNA mutants have reduced amounts of both CaM-IP2 and CaM-IP3. Western blots of flagella (8 μg) isolated from wild-type and CSC amiRNA mutants were probed with antibodies that recognize the CSC (CaM-IP2 and CaM–IP3), radial spokes (RSP1), inner dynein arm I1 (IC138), and central apparatus (PF20). CaM-IP2 amiRNA mutants 4A2 and 4D6 and the CaM-IP3 amiRNA mutants 6E6 and 1A3 have reduced amounts of both CaM-IP2 and CaM-IP3. 6E6 is the most efficient knockdown; no CaM-IP3 or CaM-IP2 is detectable by Western analysis. RSP1, IC138, and PF20 are present at near wild-type levels in each amiRNA mutant.
