Nrf2-specific shRNA lentivirus (LV-Nrf2) knock down Nrf2 protein expression. Cultures of differentiated human epithelial cells were infected at an M.O.I of 10 with shRNA lentivirus LV-Nrf2 or a GFP-scrambled control (LV-scramble). (A) NEC confocal microscopy GFP/shRNA (green) and nuclei (blue). (B) Total RNA was isolated and subjected to RT-PCR to quantify Nrf2 transcripts and normalized to the expression of β-actin. (C) Cellular lysates were harvested at 48 h postinfection and were subjected to Western blot analysis to detect Nrf2, HO-1, and β-actin. β-Actin levels were used as internal loading controls.
