Samples were either untreated or treated with phosphatase. Western (B) and northern (C) blot analysis of protein and RNA isolated from an α-factor-synchronized culture of the AP16 strain using the anti-Myc antibody (western) and probes specific for the SWI5 and RPB4 transcripts (northern). Cells were collected at 10 min intervals. The panel below the northern blot shows the fold induction of the SWI5 transcript. Samples were first quantitated relative to the RPB4 loading control then fold induction during the cell cycle was calculated using the time 0 value. The arrow in the graph indicates when maximum budding was achieved (solid arrow) following α-factor release.
