Expression of CD44 was markedly decreased after transfection with CD44 siRNA. The CD44 knockdown CD44+CD24 cells exhibited decreased expression of CD44 at a transcriptional level as confirmed by RT-PCR (A) and band intensity on agarose gel analyzed with GelAnalyzer software (E). At the translational level, this was confirmed by flow cytometry (C) and compared with cells where knockdown did not occur (B). The CD44 knockdown cells proliferated slowly with a decrease in the total number of cells compared with normal cells (D).

Notes: N1, N3, and N6 are normal cells cultured in medium supplemented with 0, 1, 3, and 6 μg/mL doxorubicin, respectively. K0, K1, K3, and K6 are knockdown cells
