Ash2LWH is a DNA-binding motif. (A) Overlay of the structures of Ash2LNTD and the FoxO1–DNA complex. Ash2LPHD is shown in cyan, Ash2LWH in yellow and FoxO1 in marine. (B) The surface area of helix α3 of Ash2LWH is highly positively charged and thus compatible with DNA binding. Ash2LNTD is in surface representation and coloured according to its electrostatic potential (positive, blue; negative, red). A concentration of 0.1 μM DNA was incubated with increasing amounts of Ash2LNTD (0.1–25.6 μM). (D) Full-length Ash2L binds with higher affinity to DNA than Ash2LNTD.
