SidL deficiency decreases the production of hyphal FC (A) and conidial HFC (B) mainly under iron-replete conditions. (A) Hyphal FC contents were measured in biological triplicates after 24 h and 48 h of liquid growth under iron-depleted (−Fe) and iron-replete (+Fe) conditions, and measurements were normalized to the wt FC content under −Fe conditions. Notably, the FC content increases with cultivation time, as shown previously for A. nidulans (7); under iron-depleted conditions, FC is accumulated in the desferri form and under iron-replete conditions in the ferri form. (B) HFC contents were determined for conidia harvested from mycelia transferred from iron-depleted (−Fe) or iron-replete (+Fe) 20-h liquid cultures and incubated for another 4 days on high-iron plates for sporulation.
