Hydrolysis of xylan substrates by five putative endoxylanases from R. albus 8. (A) Thin-layer chromatography (TLC) analysis of end products of hydrolysis. The products of reaction mixtures were resolved by TLC followed by staining with methanolic orcinol. Arabinose (A1) and xylo-oligosaccharide standards (X1 to X5) were spotted on the plate in lanes 1 and 2, respectively, to serve as markers for the identification of the released products. (B) Specific activities of the five endoxylanases. The final concentrations of the enzymes were 50 nM for reactions with soluble wheat arabinoxylan and 100 nM for oat spelt xylan and birchwood xylan as substrates. Each substrate was at 1% (wt/vol) in the reaction mixture. Bars are shown with standard errors for three independent experiments.
