Resistance analysis of BMS-346 selected substitutions. Huh-7 cells were transfected with wild-type (WT) or mutant replicon RNAs and incubated in the presence or absence of various concentrations of BMS-346. Luciferase activities were determined in lysates of cells harvested at 72 h after transfection, and EC₅₀s were determined. The n-fold resistance of the mutant RNAs relative to that of wild-type RNA (mutant EC₅₀/wild-type EC₅₀) is depicted.
