Figure 1.

Identification of epitope M133-specific T reg cells using MHC class II tetramer. (A) CNS-derived lymphocytes from rJ2.2-infected Foxp3gfp mice were analyzed for Foxp3+ CD4 T cells at the indicated times after infection (n = 5 mice for each time point, in at least two independent experiments). (B) BrdU incorporation by CNS-derived CD4 T cells at 6 d after infection is shown. Data shown are representative of six mice analyzed in three independent experiments. (C and D) Foxp3gfp mice were infected with rJ2.2 (C) or rJ2.2.MY135Q (D) and sacrificed at the indicated times after infection. Brain and spinal cord (SC)–derived lymphocytes were stained with I-Ab/M133 tetramers and representative plots gated on CD4+ T cells are shown. (E) Frequency of T reg cells that are I-Ab/M133 tetramer-positive is shown. Data are from four to seven mice per time point in two to five independent experiments. Error bars in A and E indicate mean ± SEM.
