Michaelis-Menten kinetics of Spectrozyme TH hydrolysis by human thrombin in the presence of dimer 12a. The initial rate of hydrolysis at various substrate concentrations was measured spectrophotometrically in pH 7.4 buffer as described in ‘Experimental Procedures’. The concentrations of 12a chosen for study include 0 (○), 3.0 (◆), 10.5 (▴), and 30 μM (⋄). Solid lines represent non-linear regressional fits to the data by the standard Michaelis-Menten equation to yield KM and VMAX.
