Complex 7a-induced mitochondrial permeabilization results in swelling and electrochemical gradient dissipation on isolated rat liver mitochondria. (A) Mitochondrial swelling was estimated by decrease of the relative absorbance recorded at 540 nm (rA540); (B) ΔΨ was measured by using 0.4 μM rhodamine 123 and mitochondria were energized by the addition of 5 mM succinate (suc). The arrows indicate the addition of Complex 7a, the numbers are the cyclopalladated compound concentrations at μM, and the traces are representative of at least three experiments with different mitochondrial preparations.
