PTPBR7 ectodomain does not bind to heparan sulphate and chondroitin sulphate moieties. (A) Medium containing the BR7ecto226 probe was added to microplate wells previously coated with one of the indicated GAG chains, or with coating buffer as a control. Binding was assessed by addition of pNPP as substrate and measuring absorption (ABS) at 405 nm. Successful coating of the wells was confirmed separately, exploiting GAG-specific antibodies. Intensities did not significantly differ in comparison with the uncoated control. Results (n=3) are presented as Mean ± S.D. (B) RAP in situ staining of the epidermis (arrow) in mouse embryonic cryosections using BR7ecto226 as a probe. (C) The RPTPσ ectodomain probe displays distinct binding to the basal laminae (arrow), whereas BR7ecto226 does not.
