Degradation of Atp13a2^Isoform-3. (A and B) HeLa cells were either mock transfected or transfected with the indicated myc-tagged Atp13a2^Isoform-3 expression constructs. At 20 h post-transfection, the cells were treated with MG132 (50 mm) or DMSO. The indicated proteins were detected by immunoblotting. (A) Immunoblot showing Atp13a2^Isoform-3 proteins increase after MG132 treatment. (B) Immunoprecipitation of transfected myc-tagged Atp13a2^Isoform-3 from HeLa cells after MG132 treatment. The immunoprecipitates were immunoblotted for myc and ubiquitin. Bottom panels show whole cell lysates used for the immunoprecipitations blotted for myc and ubiquitin.
