Single Amino Acid Substitutions In The D1 Domain Of KIR3DL1 Abrogates HLA Binding. (A) HEK293T cells were transfected with 2 μg of plasmid containing FLAG tagged KIR3DL1*01502, *01502 G138W, *01502 P163S and *01502 L166R and stained with PE-conjugated HLA tetramers. Dashed line represents staining with HLA-A*2402 (left panel) or HLA-B*2705 (right panel), full line represents staining with control HLA-B*0801. (B) The mean fluorescence intensity of the tetramer stained cells is expressed as fold change over the fluorescence of unstained cells.
