Schematic of templates constructed for genomic integration. Plasmids pBjk2648r-jtk2951 and pBjk2741-jtk3041 both contain: 1) a ~500 bp homology arm (HA) matching the 5' end of the genomic insertion site 2) a constitutive promoter (P_CON) 3) the ORF of the trans-acting replication factor (Trans) 4) a terminator 5) a kanamycin resistance cassette (KnR) flanked by FRT sites and 6) a ~500 bp HA matching the 3' end of the genomic insertion site.
