Inhibition of motility by chemical analogs of Q24DA. (A) Structures and IC50s. The IC50s were determined by measuring the swarm diameters produced by three independent cultures of strain C7258 stabbed into swarm agar supplemented with 2-fold dilutions of each compound (numbered 1 through 7), starting at 10 μg/ml. (B) Swarm agar test and growth in broth. Overnight cultures of strain C7258 containing >109 cells/ml were stabbed into 4 ml of soft agar containing 10 μg/ml of compounds 2 to 7. In parallel, three overnight cultures of strain C7258 were diluted 1:100 in LB broth containing the above concentration of compounds 2 to 7 in 96-well microtiter plates. Plates were incubated for 16 h at 30°C.
