Huh7.5 cells stably transduced with USP18 shRNA were treated with serially diluted IFN-α2a for 17 h. Infectious HCV (JFH) was added to an MOI of ∼0.1 and incubated for 2 days. RNA was extracted, and USP18 levels were measured and corrected for actin. Nonlinear regression analysis of the IFN-α2a dose-response curve using GraphPad Prism (version 5.01) was performed. Error bars represent the ranges of replicate data.
