SEST products accumulate in the presence of MuB. Strand transfer reactions containing the indicated ratios of DDE⁺ to DDE⁻ transposase were performed in Mg²⁺ as described in Materials and Methods. Reactions contained a 10 to 1 ratio of unlabeled to labeled substrate, so that most DEST products contained only one labeled substrate. Reaction products are labeled as in Fig. 6. The percent of strand transfer products that were single ended (%SEST) (average ± 1 s.d. of three to four experiments, calculated as in Fig. 6) is indicated below each lane. The higher level of SEST products observed in lanes 1,3–5 as compared with equivalent lanes in Fig. 6 was due to the unstaged protocol used in these experiments.
