Endoreplication from 4C to 8C is blocked in anlagen in PDD2 knockout cells. Wild-type (WT) and mutant (Δ) cells were lysed at time points indicated at right, stained with propidium iodide, and the relative DNA contents of their nuclei were measured by flow cytometry. Relative ploidy values were determined from “standards” of micronuclei purified from starved cells that are known to be in G₂ stage of the cell cycle with 4C DNA content (Allis and Dennison 1982). Peaks corresponding to micronuclei (Mi), anlagen (An), and old macronuclei (OM) are indicated.
