β2 integrin-specific neutrophil adherence in stimulated siRNA-exposed cells. Neutrophil were isolated, exposed to air or HBO2 for 45 min, and then incubated for 24 h with siRNA as described under “Experimental Procedures.” Samples labeled control in the figure were incubated with control, scrambled sequence siRNA that will not lead to specific degradation of any known cellular mRNA. After 24 h, cells were loaded with calcein AM and placed on fibrinogen-coated plates containing either PBS or PBS with fMLP or 8-Br-cGMP to achieve concentrations as shown in Table 1. After 10 min, fluid was removed, and cell adherence was calculated as described under “Experimental Procedures.” Data are mean ± S.E. (error bars), n = 4 for all studies. *, p < 0.05 versus air-exposed cells incubated with PBS after the indicated siRNA.
