Prevention of PGE₂ and NO formation reverses promastigote inhibition of IL-12 production. Cells (1 × 10⁶/well) were infected with L.mexicana promastigotes (ratio, 5:1) for 2 h and then stimulated with LPS (1 μg/ml) for 6 h. Indomethacin (30 μM) or l-NAME (1 mM) was then added and cells incubated for a further 18 h. Samples were assessed for IL-12 production as outlined in Section 2. Each value represents the mean ± S.E.M. of at least 3 experiments performed in triplicate, *** indicates p < 0.05 compared to LPS alone.
