The stability of preexisting Cx43 is altered after MMC treatment. Cells were treated with prewarmed media containing 10 μg/mL CHX with or without 5 μM MMC for indicated times in minutes. Blot shows total Cx43 using a C-terminal–specific antibody. Blots were stripped and reprobed for α-tubulin as a loading control. Quantification below is ratio of Cx43/α-tubulin normalized to the zero time (0) for “CHX” or “CHX + MMC.” Asterisks indicate statistically significant differences between “CHX” and “CHX + MMC” (2-way ANOVA, P < 0.01; Bonferroni multiple comparisons, P < 0.05).
