HCRLC-PAGFP reconstituted fiber under p-polarized TIR illumination. Image was taken with a 3 s exposure time using a 100×, 1.49 NA objective and 16 × 16 μm pixel EMCCD camera. Fluorescence shows the striated pattern of the myosin thick filaments. Single molecules are isolated bright spots of photoactivated PAGFP. Background comes from unphotoactivated PAGFP.
