EphB2-Fc dephosphorylates PAG/Cbp in a γ-secretase-dependent manner. A) Rat primary neurons at 14 DIV were stimulated with Fc or EphB2-Fc for 10 min in the presence or absence of γ-secretase inhibitor L685,458. Cell extracts were immunoprecipitated with anti-PAG/Cbp antibody and probed by WB for phosphotyrosine (top panel) or PAG/Cbp (bottom panel). B) HEK293 cells stably transduced with ephrinB2 were transfected with PAG/Cbp and stimulated with Fc or EphB2-Fc for 10 min in the presence or absence of γ-secretase inhibitor L685,458 as in A. EphB2-Fc promotes the tyrosine dephosphorylation of PAG/Cbp in both neurons and HEK293 cells, and this is inhibited by γ-secretase inhibitor.
