Left panel: HEK293 cells stably transduced with PAG/Cbp were treated with Fc or EphB2-Fc for 10 min in the presence or absence of γ-secretase inhibitor L685,458. Cell extracts were immunoprecipitated with anti-Csk antibody and probed by WB for PAG/Cbp (top panel) or Csk (middle panel). Bottom panel (input) shows levels of PAG/Cbp. EphB2-Fc promotes the dissociation of Csk from PAG/Cbp, and this is inhibited by the γ-secretase inhibitor. Right panel: densitometric quantification of EphB2-induced decrease in PAG/Cbp-Csk complex. Csk levels in EphB2 treated-cells were normalized to Csk levels in Fc-treated cells. Bars represent means ± se of 3 independent experiments.
