The dependence of the yield on non-specific nucleosomes on the concentration of NaCl. The solution of nucleosomes (0.4 nM) was incubated in the buffer solution (10 mM HEPES, 4 mM MgCl₂) in the presence of 1.6 mM of CHAPS and various concentrations of NaCl for 2 hours. The nonspecific and specific complexes were identified by the measurements of the lengths of the arms on the AFM images. Nucleosomes with a short arm less than 10 nm were assigned to the class of non-specific nucleosomes.
