A. IVIS 100 imaging system was used to monitor tumor growth in real-time. We found that tumor growth is inhibited dramatically with the treatment of 1D11 compared with Group 2 (13C4 treatment) and 3 (No treatment control). B. 1D11 treatment inhibits the tumor growth in a time dependent manner. C, At the end of the 45-day treatment period, mice were sacrificed and tumors were isolated. The average tumor weight and volume was 5.3 g and 6.85 cm3, respectively in 1D11 treatment group. In comparison, the average weight and volume of tumors in the 10 animals treated with the control 13C4 was significantly greater at 15.8 g and 12.85 cm3, respectively. The corresponding values in the mice that received no treatment were 31.4 g and 23.39 cm3, respectively (P<0.01 among three groups). D. Immunohistochemical analyses of the primary tumors revealed that the expression of p-ERK, DNMTs in animals with 1D11 treatment is significantly lower than those of the other two groups.
