HDA2 and HDA3 are required for the in vitro deacetylase activity of the HDA1 complex. α-HDA1 antibody was used to immunoprecipitate histone deacetylase activity from whole-cell extracts. Yeast strains YDS2 (WT), WJY110 (hda1Δ), WJY103 (hda2Δ), WJY104 (hda3Δ), and WJY105(hda2Δhda3Δ) were used. Half of each precipitate was used to assay the enzymatic activity (A) and half to determine the amount of the HDA1 protein by Western blot (B).
