Hydroxyl radical cleavage of the RT1t49(−5) variant RT1ZM bound to RT. The patterns of cleavage from representative hydroxyl radical digestions are shown. RT1ZM is subject to hypersensitive cleavage at position T12 when in complex with wt HXB2 RT but not when it is in complex with the RNase H defective E478Q mutant. Hypersensitive cleavage at position 12 is enhanced in the absence of EDTA (inset), which competes with RT for free Fe(II).
