Characterization of PilA expression in different F. tularensis backgrounds using immunoblotting of whole-cell lysates with anti-PilA antibodies. From left to right, the lanes contain strains FSC200 (wild type [wt]), FSC679 (FSC200 ΔFTL_0425pglA]), FSC749 (FSC200 pilAFLAG), FSC703 (FSC200 pilAFLAG ΔFTL_0425 [pglA]), SCHU S4 (wt), and PS1 (SCHU S4 pilAFLAG). Note that the presence of FLAG-tagged residues decreases the mobility of PilA and that in the SCHU S4 background (strain PS1 in the rightmost lane), it may alter posttranslational modification of PilA.
