The effect of MEKK2 gene disruption on MKK7 activation in response to cross-linking of FcɛRI to KL and IL-3 in ESMCs. Passively sensitized MEKK2+/+ and MEKK2−/− ESMCs were stimulated with OVA for 10 min, and MKK7 activity was measured as described in Materials and Methods. Otherwise, MEKK2+/+ and MEKK2−/− ESMCs were stimulated with 100 ng/ml KL or 50 ng/ml IL-3 for 10 min, and MKK7 activity was measured. MKK7 protein levels were determined by Western blot analysis. Representative autoradiographs from three independent experiments are shown.
