Expression of kinase-inactive MKK7 does not affect promoter activity of TNF-α. MC/9 cells (5 × 106 cells) were transfected with 2.5 μg pGL3TNF together with 7.5 μg MKK7K63R or 7.5 μg MKK4K116R or an equivalent amount of empty vector (pCMV5, pLNCx). The transfected cells were passively sensitized with anti-OVA IgE for 16 h and washed three times and incubated for an additional 8 h with 10 μg/ml OVA (filled bars) or PBS (empty bars).
